Postdoctoral Research Associate-Tumor Cell Biology   A Postdoctoral Research Associate position is open in the Li laboratory to study the dysregulated transcription dependency under the 3D chromatin niche in human cancers. Cutting-edge technologies and multidisciplinary approaches, including genetic and epigenomic CRISPR editing, comprehensive genetic screens, and the auxin-induced protein degradation system, are used to investigate the precise function of transcription factors and noncoding segments involved in gene transcription, chromatin architecture, and chromatin accessibility maintenance in pediatric leukemia. Those efforts are anticipated to shed light on understanding how gene regulation is precisely controlled in temporal and spatial gene expression in cancers and will provide insights into alternative therapy development. Postdoctoral Associate Position in Structural Biology    The Mittag laboratory invites applications for a Postdoctoral Research Fellow to work on in vitro biophysics studies of the mechanism of stress granule aging and its role in the pathology of neurodegenerative diseases. Recent work (from our lab and others, see e.g., Molliex et al. Cell 163: 123-133, 2015) has revealed that phase separation can promote aggregation of RNA-binding proteins. Solid deposits of these proteins are the pathophysiological hallmark of ALS and related neurodegenerative diseases. The mechanism by which phase separation enhances aggregation or promotes dynamic arrest of dense phases is unclear and will be the focus of this project. This project is part of a collaboration with Drs. J. Paul Taylor (St. Jude, HHMI) and Dr. Rohit Papu (Washington University in St. Louis).   The successful candidate will join a highly collaborative laboratory focused on elucidating mechanisms and function of biomolecular condensates in the Department of Structural Biology, with access to state-of-the-art instrumentation and expertise in NMR spectroscopy, X-ray crystallography, cryo-EM, single molecule techniques, protein technology, and microscopy. A bioinformatics postdoctoral position is available in the Savic laboratory to study the pharmacogenomics of treatment in childhood leukemia. The primary research focus of the Savic laboratory involves studying the gene regulatory architecture of pediatric leukemia to define how the noncoding portion of the human genome impacts chemotherapeutic drug response, chemotherapeutic drug resistance and leukemia relapse. To address these critical questions, the Savic laboratory applies various orthogonal functional genomic assays and high-throughput screening approaches to identify and functionally characterize genetically and/or epigenetically disrupted cis-regulatory elements impacting childhood leukemia pharmacogenomics. The long-term goal of the Savic laboratory is to gain a better understanding of the genetic and epigenetic underpinnings of chemotherapeutic drug resistance and relapse in childhood leukemia.

A wet-lab postdoctoral position is available immediately in the Northcott Lab (Department of Developmental Neurobiology) to join a highly collaborative team studying the developmental and molecular basis of the childhood brain tumor medulloblastoma. The Northcott Lab has established an internationally renowned track record in pediatric neuro-oncology, substantiated by numerous impactful publications in Nature, Cancer Cell, Lancet Oncology, and Journal of Clinical Oncology. Leveraging our exquisite access to patient material linked to large clinical trial cohorts, we are applying a combination of innovative, multi-omic approaches to discover new biological, mechanistic, and clinical insights responsible for medulloblastoma initiation, progression, and relapse.   The successful applicant will be part of a multi-disciplinary team studying the biological and clinical utility of medulloblastoma liquid biopsies obtained from unprecedented clinical trial cohorts (Liu, Smith, Kumar et al, Cancer Cell, 2021). Experience in cancer biology, molecular and cellular biology, genomics/epigenomics/transcriptomics, and clinical sample processing are highly desirable. Individuals possessing hands-on experience with sample processing for next-generation sequencing, single-cell genomics, and/or liquid biopsies will be deemed highly competitive. Dr. Zhaoming Wang, a joint faculty member between St. Jude Department of Epidemiology and Cancer Control and the Department of Computational Biology, is seeking highly motivated and creative candidates for a fully supported postdoctoral fellow position to conduct high impact clinical research primarily in genetic epidemiology of cancers (childhood and subsequent adulthood cancers), biomarker discovery for treatment- and/or aging-related late effects of survivors of childhood cancer, and omics-based precision preventive medicine.   You will have the opportunity to work with large-scale whole-genome sequencing, whole-exome sequencing, epigenetic profile, and RNA sequencing data for a well-established cohort of childhood cancer survivors with clinically assessed rich set of phenotypes including subsequent neoplasms.   In this position, you will gain extensive experience and training in genetic and molecular epidemiology, as well as programming skills and analytic expertise in computational biology. We are especially interested in understanding the roles of linear energy transfer and treatment plan robustness.   Epigenetic Characterization and Therapeutic Discovery in Pediatric Brain Tumors   Seeking a highly motivated, organized, and responsible individual to lead a major component of our new cancer genetics and epigenetics lab, as a foundation for their career advancement in academia or industry. Candidates will leverage cancer genomics and epigenomics data that we have generated in a variety of pediatric and adult brain tumors to undertake experiments in functional discovery, validation, as well as the development of integrated large-scale datasets using new genetic technologies. Successful candidates will lead research projects focusing on structural mechanisms of membrane proteins and protein machineries.  We use cryo-EM, biophysical, electrophysiological and biochemical approaches to study the function of membrane proteins and protein complexes (Sun, NSMB 2020; Sun and Mackinnon, Cell 2020; Sun and MacKinnon, Cell 2017). The lab has fully access to the state-of-the-art cryo-EM facility in St. Jude, which houses a 300kV Titan Krios and a 200 kV Talos Arctica electron microscopes, both equipped with K3 detectors. Postdoctoral Position in Radiation Oncology and Chemical Biology A unique postdoctoral fellow position is available immediately in the joint laboratory of Dr. Christopher Tinkle, Department of Radiation Oncology, and Dr. Anang Shelat, Department of Chemical Biology and Therapeutics, to apply state-of-the-art chemical biology and bioinformatics methodologies across diverse pediatric malignancies to investigate the molecular consequences of defective DNA repair, and to target these deficits using small molecules. Postdoctoral Research Associate in Immunology and Immunometabolism in Dr. Hongbo Chi’s Laboratory   Immunology postdoctoral positions are available in the laboratory of Dr. Hongbo Chi in the Department of Immunology, St. Jude Children’s Research Hospital (www.stjude.org/chi), to investigate discovery and cancer immunology, by focusing on cell metabolism of the immune system (immunometabolism) and its implications in cancer and other diseases. As metabolic pathways are inextricably connected to immunity and cancer, understanding these connections allows us to gain new insights into biological concepts and disease targets. We are particularly interested in understanding the metabolic programs (e.g. glucose, lipid, and mitochondrial metabolism), signaling pathways (e.g. mTOR and Hippo kinases; metabolite- and nutrient-induced signaling), and systems-level regulatory networks (e.g. interactions between protein regulators and their targets) in basic T cell and dendritic cell biology and antitumor immunity. Postdoctoral Research Associate   Overview We are recruiting a Postdoctoral Research Associate (a Cell and Molecular Biologist) to join the efforts in chemically modulating transcription factors. The Chen Lab (https://www.stjude.org/research/labs/chen-lab-taosheng.html) uses multidisciplinary approaches to study transcriptional regulation by nuclear receptors, including chemical biology approaches to develop chemical modulators, with a goal to understand nuclear receptor-regulated transcription networks and design therapeutic approaches to overcome drug resistance and tumorigenesis in cellular and animal models. The cell and molecular biology efforts are expected to synergize with our pharmacology, structural biology, and medicinal chemistry efforts and enable publication of impactful papers (e.g., Nat Commun 8:741, 2017; J Am Chem Soc 143:18467, 2021; Nucleic Acids Res 50:3254, 2022). Changes in the Structure and Organization of the Genome During Neurogenesis   During retinal development, more than 8,000 genes change in their expression as multipotent retinal progenitor cells produce each of the 7 classes of cell types in an evolutionarily conserved birth order. Although it has been well established that multiple changes in the epigenome accompany changes in gene expression, little is known about how those processes are coordinated during neurogenesis. Over the past 5 years, we developed a detailed map of the structure and accessibility of the human and mouse retinal genome during development. Specifically, we performed a multifaceted integrated analysis that included profiling of the covalent modifications to the DNA and histones, promoter structure, chromatin accessibility, looping interactions, and euchromatin/heterochromatin localization (https://pecan.stjude.cloud/retinalnucleome). One of the most significant discoveries to come from this comprehensive analysis was the identification of a series of core regulatory circuit super-enhancers (CRC-SEs) adjacent to genes having important roles in retinal development, including Vsx2, Crx, Six3, Otx2, Fgf15, and Ascl1. Each of those CRC-SEs have been deleted in mice using CRISPR-Cas9 and analysis is ongoing. We are also analyzing the evolutionary conservation of those CRC-SEs in human stem cell derived retinal organoids. The results of these studies will be important for filling a fundamental gap in our knowledge about the role of CRC-SEs in retinal development and will set the stage for characterization of CRC-SEs in other genes required for retinogenesis. We are seeking a highly motivated postdoctoral fellow to study the role of CRC-SEs in retinal development and diseases such as macular degeneration and diabetic retinopathy. A fully funded postdoctoral position is available in the Clements group in the Department of Hematology at St. Jude Children’s Research Hospital to study how the adult hematopoietic system is established during vertebrate embryonic development. We are interested in understanding how early precursors of the sympathetic nervous system and vascular smooth muscle precursors interact with developing endothelial cells to establish the earliest hematopoietic stem cells. Our recent findings (Nature Cell Biology, 19:457-467, 2017) and unpublished data define the existence of this connection in in vertebrates and we are now working to better understand key details. In addition to a Ph.D., applicants should have a record of publication, and experience with molecular, biochemical, and cell biological techniques.
